Protocols

In situ hybridization of whole-mount larval zebrafish. This protocol extends standard embryo in situ protocols for use at metamorphic stages (10–28 dpf). The protocol is still under development and is updated periodically. Last update: 26 June 2007.
In situ hybridization on sections of larval zebrafish. For detecting mRNA on cryosections of larval zebrafish.
Clearing and staining for bone and cartilage in larval fish. An alcian blue and alizarin red clearing and staining procedure modifed for use with larval zebrafish.
Gateway and Tol2 transgenesis. A protocol for using Gateway vectors and the Tol2 transposable element system for generating transgenic fish.

Development time – temperature converter. There is a static pdf version and a dynamic Excel version. For each column, you can change the temperature by changing the top cell and you can put in a specific number of hours or days by changing the left columns, or by typing in the white cells at the upper left. You can also use this to estimate hours until a particular stage is reached (e.g., 5 hours to go at 28.5 C is equivalent to 6.7 hours to go at 24 C). The table is based on Chuck Kimmel's estimated linear function in embryos and may or may not be accurate at later stages.
